Primary Cell Development

Description: Primary cell cultures are established from a specific organ site with specific processing. Primary cell cultures "mimic" in vivo conditions more closely than cell lines.

Cell Maintenance

Description: The cell cultures are monitored 3-5 times per week. Cell cultures will be maintained at an appropriate density in specific media. The cells (monolayer or suspension) will be sub-cultured based on standard methods.

Cell Expansion

Description: Cell cultures are maintained (primary or established lines) in log phase to generate a predetermined number of cells. Useful for generating the number of cells needed for experiments such as protein purification, DNA isolation, creating master cell banks and working cell banks.

Cell Line Banks: Master Cell Banks & Working Cell Banks

Description: Cells will be amplified in culture to create a master cell bank (MCB) of 20 cryovials of cells in log phase at one passage number. Amplification of the cell line will be continued until enough cells are available at the same passage number for a working cell bank (WCB) consisting of 200 cryovials.

Cryopreservation

Description: Cells in log phase will be cryo-preserved at 10⁶ cells/ml in CPZ^TM. Cryopreservation allows cell lines to be banked for further experiments at a later date. Also, cryopreservation can be a guard against accidental loss of cells in culture due to contamination or mechanical difficulties.

Immortalization

Cell Cloning

Description: Cells are cloned by limiting dilution. Cell cloning (96 well plate) and transfer to a 24 well isolates and preserves a specific cell type and its specialized properties.
